I just tried and was able to activate content in my dev org. I'm a newbie and it took me a while to figure it out.
First, enable Content:
App Setup > Customize > Salesforce CRM Content > Check enable Content Checkbox
Then set user permission:
Admin Setup > Manage Users > Users > Edit User > Check Salesforce CRM Content User Checkbox

No - Salesforce Content is not available in DE. You can sign-up for a trial of Content here.

Anyone manage to enable it?
Hey.. I tried with an old deveoper org (was created around 4-5 yrs back). In there I cannot enable Salesforce Content. but I tried it on a new developer org (created 1 month back). I was able to activate Salesforce Content successfully.
I had the same experience. On older DE sandbox it wouldn't activate. On newer DE sandbox it did.
