Assignment Rules and Lead Insertion via API

If we are inserting leads into Salesforce via the API, do Assignment Rules still trigger?

not by default you can use the AssignmentRuleHeader to have the rules fired.

Yes, they will be executed if the record is created manually, captured from the web, or imported via the lead import wizards. Here's an example of ensuring execution with Data Loader (which uses the API).
