Salesforce1 app Users

Is there a way to know in a Org how many users are using Salesforce1 App

Sure, this is possible 

if you are the Admin, pls go to Setup > Adminsiter > Manage Apps > Connected Apps OAuth Usage > Salesforce1 for iOS > you will see the user Count against the application you wish to check - in your case it will be Salesforce1
